Breast cancer patients whose tumors overexpress HER2 and/or express HER2-receptor are more likely to develop a more aggressive disease. Clinical studies have shown that statins exert a chemopreventive effect on different types of malignancies. This study aimed to evaluate the synergistic effect of the atorvastatin on trastuzumab in HER2-positive breast cancer. In this study, five HER2-positive breast cancer cell lines, SK-BR-3, BT-474, MCF-7, ZR-75-30, and SK-BR-3 cells, were incubated in the presence of increasing concentrations of atorvastatin or trastuzumab for 24 hours. Cytotoxicity was determined by trypan blue exclusion. Cell cycle analysis was performed by fluorescence-activated cell sorting. Cell migration was evaluated by wound healing assay. Combination index analysis was used to evaluate the dose-reduction index (DRI) and isobologram analysis to analyze the type of interaction. In SK-BR-3, MCF-7, and ZR-75-30 cells, the combination of trastuzumab and atorvastatin reduced the IC50 values to a greater extent than either drug alone. Atorvastatin and trastuzumab induced G1 phase arrest. The combination of trastuzumab and atorvastatin inhibited SK-BR-3 cell migration to a greater extent than the trastuzumab or atorvastatin alone. In MCF-7 and ZR-75-30 cells, combination of trastuzumab and atorvastatin did not result in a synergistic interaction. In addition, the combination of atorvastatin and trastuzumab reduced the number of SK-BR-3-derived colonies to a greater extent than either drug alone. These be359ba680
